AIM

To extract and visualize DNA from an intestinal bacteria in exponential growth phase.

To establish the existence of matrilineal kinship between a son and two possible biological mothers by analysing part of the HVII hypervariable region of the mitochondrial DNA of the three individuals.

To analyse whether a Genetically Modified Organism (GMO) is maize or soya by studying the zein and lectin genes, respectively.

To do a qualitative analysis of the Cauliflower Mosaic Virus 35S promoter and the Agrobacterium tumefaciens NOS gene.

LEVEL

Can be adapted to O-level, A/AS-level, Science colleges and Universities.

TECHNIQUES

Inoculation and incubation of microorganisms in the culture media that is specific for their growth.
Extraction and visualization of DNA processes.

DNA electrophoretic analysis of each sample, which has been previously amplified by PCR.
Staining and visualization of DNA bands.

Reading and comparison of different mtDNA sequences.

DURATION

Practice sessions can be overlapped to save time. Please, check the duration of each individual practice.

CONTENT

Each kit contains four work stations with all the necessary material and components to perform the experiments: racks, microorganisms, culture media, lysis solution, precipitation solution, PCR amplified mitochondrial DNA samples for each individual, DNA for the positive and negative control, PCR amplified DNA samples for each of the gene fragments (each group is assigned four genes of a different variety of food), agarose gels, patented equipment for “easy” loading of gels, electrophoresis and staining solutions and printed mtDNA sequences.
